It would have worked but Sisyphus had already started to form into a prime soul by the time the angels imprisoned his soul, so if Sisyphus was awakened, he maybe could break free. That's why P-2 is so heavily guarded, first you need to P-Rank every level in the Prelude and Act 1, then beat Minos Prime, then P-Rank Act 2, then get through the absolute hell that is P-2, and only then you get to fight at the Flesh Panopticon.
